Transport in Living Things

Introduction:

-For living organisms to stay alive, chemicals must be moved easily:

(a) From one part of a cell to another (intracellular)
(b) Into and out of a cell
(c) From one cell to another

Chemicals – oxygen, nutrients, minerals, waste, carbon dioxide etc

Subtances must be soluble before they can move in and out of cells and across the plasma membrane (partially / selective permeable)

-The 3 process of material exchange are:
(a) Diffusion
(b) Osmosis
(c) Active transport

Diffusion
-Definition – Movement of particles from a region of their higher concentration to a region of their lower concentration, down a concentration gradient.
-A passive process; it is non-energy requiring
-Diffusion occurs in gases, liquids or solids and it occurs in the presence or absence of a partially permeable membrane
-A concentration gradient or a diffusion gradient refers to the difference in the concentration of the molecules between the 2 regions
-The permeable membrane allows both sugar and water molecules to pass through it.
-Once the concentrations of both compartments are equal, no more net movement of particles will occur.
-Factors that affect rates of diffusion are:
(a) Concentration Gradient - Greater gradient -> faster rate; e.g. constant blood flow, constant gradient maintenance
(b) The distance that the particles have to travel – thinner -> faster rate; Some organisms depend on diffusion to meet physiological needs -> adaptation in structure; e.g. Flattened body (leaves, flatworms)
(c) Surface area : volume ratio – greater SA : volume ratio -> faster rate SA/V; e.g. Nutrients enter cells in the small intestine after digestion
(d) Size of particles – Small mass -> faster rates
(e) Nature of structure across which diffusion occurs – The greater the number and size of pores incell membrane -> greater rate of diffusion
(f) Environmental conditions – Increase in temperature or pressure -> faster rate of diffusion

Osmosis
-Net movement of free water molecules from a region of higher water potential to a region of lower water potential through a partially permeable membrane
-Osmosis is a special form of diffusion:
-Involves only water molecules
-A partially permeable membrane must be present

Water potential:
-Water potential of a solution is a measure of whether it is likely to gain or lose water molecules from another solution
-Greater concentration (higher proportion) of solutes – lower water potential
-Lower concentration (lower proportion) of solutes – higher water potential
-Water diffuses passively from the less concentrated solution into the more concentrated solution through the partially permeable membrane.
-Pure water has the highest water potential, which is zero.
-A dilute solution contains more water molecules per unit volume than a concentrated solution, so it has a higher (less negative) water potential than a concentrated solution which has lower (more negative) water potential.

Importance of water potential gradient in the uptake of water by plants
-Plants absorb water from the soil through the root hairs.
-The soil solution is usually less concentrated (higher water potential) than the cell sap in the vacuole of the root hair (lower water potential)
-Water moves into the root hair cell by osmosis down a water potential gradient through a partially permeable membrane (plasma membrane of the root hair cell)
-Water passes from one cell to another by osmosis until it reaches the xylem vessels.
-The root hair cell is adapted to absorption by having a long and narrow extension which increases the surface area : volume ratio thereby enhancing the rate of absorption of water (osmosis) and mineral salts (active transport).

-A solution is considered hypertonic to a 2nd solution when it is more concentrated than the 2nd solution.
-A solution is considered hypotonic to a 2nd solution when it is less concentrated that the 2nd solution.
-Solutions are considered to be isotonic to each other when they are of the same solution.

Effect of osmosis on plant and animal cells
The effect of osmosis on animal cells
-Animal cells are placed in distilled water (hypotonic solution) => Water enters cells by osmosis -> Cells swell and burst -> Plasma membrane ruptures
-Animal cells are placed in concentrated salt solution (hypertonic solution) => Water leaves cells by osmosis -> Cells shrink and shiver -> Crenation occurs

The effect of osmosis on plant cells
-Plant cells are placed in distilled water (higher water potential) => water enter cells by osmosis -> Cells enlarge are becomes turgid.
-Plant cells are placed in a concentrated salt solution (lower water potential) that is more concentrated than the cell sap of the vacuole => Water leaves cells by osmosis -> Cell body shrinks and pulls away from cell wall -> Cell decreases in size and becomes flaccid. The cell contents pull away from the cell wall (plasmolysis)

Active Transport
-Transport of substances into and out of the cell(s) against a concentration gradient with the expenditure of energy.
-The energy (ATP: Adenosine Triphosphate) required by the cell is provided by the chemical reaction of respiration.
-Examples:
(i) Root hair cells take in nitrate ions from the soil although the concentration of nitrate ions inside the root hair cells is higher that the concentration in the soil.
(ii) Glucose and amino acids are actively transported from the lumen of the intestine into the cells of the vili.

Transport Systems
-In a one-celled organism called Amoeba, oxygen diffuses in and carbon dioxide diffuses out of it efficiently
-However, there is a need for a transport system in multicellular organism due to:
(a) Exchange of materials
~ Supply of oxygen and nutrients
~ Removal of carbon dioxide and waste products
(b) Limitation of simple diffusion
(c) Low SA to volume ratio

Transport system in plants
-2 types of transport tissues; xylem and phloem
-Xylem – transports water and dissolved mineral salts from roots and stem to leaves
-Phloem – transports nutrients from leaves and to the rest of plant.
-Water and dissolved mineral salts move from the cells at the outer layer into the inner cells until they reach the xylem vessels
-How water moves up the xylem vessels
~The evaporation of water from the leaves results in a suction force which pulls water up the xylem vessels ~This suction force is called transpiration pull
~The stream of water up the plant is called transpiration stream

Animal transport system (Circulatory system):
-Human circulatory system consists of
(a) The heart
(b) Blood vessels
(c) Blood

~Oxygenated blood, blood travels from
Lungs -> pulmonary veins -> left atrium -> left ventrical -> aorta -> rest of body
~Deoxygenated blood, blood travels from
All parts of the body -> vena cava -> right atrium -> right ventrical -> pulmonary artery -> lungs

(b) Blood vessels (i) Arteries
~Carries oxygenated blood away from the heart (except pulmonary artery)
~Thick muscular walls; Small lumen
~No lunar valves
(ii) Veins
~Returns deoxygenated blood to the heart (except pulmonary vein)
~Thin walls; large lumen
~Semi-lunar valves present to prevent backflow of blood
(iii) Capillaries
~Exchange materials between blood and tissue cells through wall.
~Only one cell thick
~No semi-lunar valves

(c) Blood
(i) Red blood cells
~contain a red pigment, hemoglobin which carries oxygen from the lungs to all cells in the body
~each red blood cell has a biconcave shape which increases surface area : volume ratio for diffusion of oxygen
(ii) White blood cells
~ Involved in the body’s immunity through phagocytosis (engulfing bacteria) and antibody-production
(iii) Platelets
~Necessary for blood clotting
(iv) Plasma
~ Contains water, proteins, mineral salts, carbon dioxide, digested food, waste products and hormones

Allimentary Canal: - Mouth
1. Teeth - Breaks the food up into smaller pieces to increase the surface area for the enzymes to act on.
2. Tongue - rolls the food into small spherical masses (boli) for swallowing
3. Saliva - contains water to moisten food or to dissolve chemicals in the food so that they can reach the taste buds, mucin to lubricate the food, salivary amylase which catalyses the breakdown of starch to form maltose

Covers the trachea during swallowing

-Oesophagus
Peristalsis - a series of coordinated rhythmic muscle contractions that propel food to the stomach

- Stomach
1. Muscular walls - churns and breaks up the food particles
2. Gastric juice - contains HCl and pepsin (protease). Pepsin enzymes catalyses the break down of protein into polypeptides. HCl acidify the environment in the stomach for the actions of pepsin and to kill potentially harmful bacteria in the food.
3. Pyloric sphincter - After digestion in the stomach, the pyloric sphincter will relax to allow the food to enter the duodenum

Procedure: Add a few drops of Benedict’s solution to a solution of the
substance to be tested and heat in water at 100°C for 5 mins.
Positive reaction - the blue Benedict’s solution formed a brick-red precipitate in the presence of reducing sugar
Negative reaction- The blue Benedict’s solution remains blue in colour.

Biuret test for proteins:
Procedure: Add 2 ml of sodium hydroxide solution to the substance to be tested, then add a few drops of copper sulphate solution (1%) to the mixture
Positive reaction- the sample turns violet in colour
Negative reaction- the sample turns blue in colour

Grease spot test for oil:
Procedure: Drop a sample of the substance to be tested on a piece of paper
Positive reaction- A permanent transparent spot is observed on the paper
Negative reaction – A wet spot is observed on the paper which dries to leave no permanent spot

Ethanol emulsion test for fats:
Procedure: Add a few drops of the substance to be tested to 2 ml of water in a test tube. Shake the mixture and add 1 ml of ethanol in the test tube
Positive reaction- a white cloudy emulsion is observed
Negative reaction- No white cloudy emulsion Is observed. The solution remains clear
